چکیده

The killifish Nothobranchius furzeri is the shortest-lived vertebrate that can be bred in the laboratory. Its rapid growth, early sexual maturation, fast aging, and arrested embryonic development (diapause) make it an attractive model organism in biomedical research. Here, we report a draft sequence of its genome that allowed us to uncover an intra-species Y chromosome polymorphism representing-in real time-different stages of sex chromosome formation that display features of early mammalian XY evolution "in action." Our data suggest that gdf6Y, encoding a TGF-β family growth factor, is the master sex-determining gene in N. furzeri. Moreover, we observed genomic clustering of aging-related genes, identified genes under positive selection, and revealed significant similarities of gene expression profiles between diapause and aging, particularly for genes controlling cell cycle and translation. The annotated genome sequence is provided as an online resource (http://www.nothobranchius.info/NFINgb).

The African Turquoise Killifish Genome Provides Insights into Evolution and Genetic Architecture of Lifespan

Lifespan is a remarkably diverse trait ranging from a few days to several hundred years in nature, but the mechanisms underlying the evolution of lifespan differences remain elusive. Here we de novo assemble a reference genome for the naturally short-lived African turquoise killifish, providing a unique resource for comparative and experimental genomics. The identification of genes under positi...

Karyotypic Study and Chromosome Evolution in Some Iranian Local Onion Populations

Abstract A karyotypic study was performed on 12 Iranian local onion (Allium cepa L.) populations. A number of mitotic cells at metaphase stage for each population were prepared. Chromosomes of suitable mitotic cells were counted and various parameters, including long arm (L), short arm (S), total length of chromosome (TL), relative length of chromosome (RL), arm ratio (AR), r-value, total chro...

Nothobranchius furzeri: A Model for Aging Research and More.

The short-lived killifish Nothobranchius furzeri inhabits ephemeral ponds in southeastern Africa and is characterized by rapid growth and early sexual maturation. With respect to the molecular, cellular, and integrative traits of aging, N. furzeri shows significant similarities to mammals, including humans. Recently, reference sequences for the N. furzeri genome have been published. Also, metho...

Mapping loci associated with tail color and sex determination in the short-lived fish Nothobranchius furzeri.

The African fish Nothobranchius furzeri is the shortest-lived vertebrate species that can reproduce in captivity, with a median life span of 9-11 weeks for the shortest-lived strain. Natural populations of N. furzeri display differences in life span, aging biomarkers, behavior, and color, which make N. furzeri a unique vertebrate system for studying the genetic basis of these traits. We mapped ...

I-38: Chromosome Instability in The Cleavage Stage Embryo

Recently, we demonstrated chromosome instability (CIN) in human cleavage stage embryogenesis following in vitro fertilization (IVF). CIN not necessarily undermines normal human development (i.e. when remaining normal diploid blastomeres develop the embryo proper), however it can spark a spectrum of conditions, including loss of conception, genetic disease and genetic variation development. To s...
